If this doesn’t help, try to move your Steam folder out of the Program(x86). All you have to do is to Right-click on Steam and LOOT and run them as Administrator. This folder has native protections and sometimes LOOT can not access it. This happens when they both are in the Program(x86) folder. Sometimes this error occurs when your steam and Skyrim cannot be accessed. Solution 2: Run LOOT and Steam as Administrator Launching the game from steam will help register the files that LOOT needs to detect. When you launch Skyrim from Steam, wait for the splash menu load before exiting. If your LOOt can not detect Skyrim then the first thing you can do is to launch it through Steam.
